Acclimatization and field adaptation of in vitro cultured Vigna luteola SC-123

Abstract
The legume Vigna luteola SC-123 has been recommended by the genebank of Sancti Spíritus Experimental Station for Pastures and Forages considering their promissory behavior, once defined the tissue culture propagation of this specie is objective of the present work to study the plantlet acclimatization and adaptation to field conditions. Three substrates were proven: manure, compost and a mixture of soil and worm humus (7:3), as well as the stimulation by a solution of naftalenacetic acid (NAA, 0.01%) for immersion of the roots. On the field several plantlet distances were proven (0.60 x 0.20 m, 0.60 x 0.60 m and 0.60 x 1.00 m), and it was compared with the traditional method used for seed production. The mixture of soil and worm humus obtained the best results for the evaluated variables, and the application of NAA solution impacted significantly in plantlet survival during the acclimatization to ex vitro conditions. The field adaptation showed indexes of survival superiors to 96% for all the proven treatments.
